Pharmacokinetics

It is poorly absorbed from the gut. It is split in the colon into 5-aminosalycilic acid and sulfapyridine by colonic bacteria. They are metabolised in the liver by aceylation at N-4 by the non-microsomal enzymes. They are partly excreted as the unchanged drug and partly as metabolic products in the urine. They are excreted mainly by the kidneys through glomerular filtration.

Indications

It is used to treat Ulcertive colitis and Crohn's disease.

Routes of Administration and Dosage

Dose: Adults;1-2 g q.i.d for 2-3 weeks, maintenance 2 g/day . Children; 40-60 mg/kg/day, maintenance: 20-30 mg/kg/day in 4 divided doses.

Contra Indications

Sulfasalazine is contraindicated in hypersensitivity to the drug,intestinal obstruction, urinary obstruction, SLE and blood dyscrasias.

Precautions

Precaution to be taken in hepatic damage, renal damage, diabetes, pregnancy and lactation.

Interactions

Reduces the serum levels of digoxin.Interferes with absorption of folic acid.
