Pharmacokinetics

Absorption of tetracycline varies with the type of tetracyline used. Food affects its absorption. Tetracycline gets widely distributed in the body and is mainly concentrated in the liver, spleen, bone marrow, and in sputum. It is excreted through kidneys via glomerular filtration.Plasma half life is 6 to 10 hrs.

Indications

It is bacteriostatic in action, and acts against broad spectrum of Gram positive and Gram negative organisms in addition to other organisms like Rickettsia, Mycoplasma, Chlamydia.

Routes of Administration and Dosage

Oral dose ; 250 to 500 mgs qid or tds .

Contra Indications

Contraindicated in children under 12yrs of age, SLE, pregnancy, lactation, and in hypersensitivity.

Precautions

Patients with hepatic and renal disease may have an increased chance of adverse effects if they use tetracycline.

Interactions

Interaction may occur with Antacids, Calcium supplements, Cholestyramine, Choline and magnesium salicylates, Colestipol, Iron-containing drugs, Laxatives, Magnesium salicylate: these drugs may decrease the potency of tetracyclines. Oral contraceptives containing estrogen: tetracyclines may decrease the contraceptive effect and increase the chance of unwanted pregnancy.
